Piracetam is indicated for the treatment of memory disorders, dementia, in Parkison’s disease, alcoholism, stroke, age related memory loss, head injury & adjunctive treatment in cortical myoclonus origin.
Dosage varies considerably according to the indication, age, formulation and renal function.
Piracetam is predominantly eliminated by the kidneys; therefore, dose adjustment is required in renal impairment.
Usual dosage for adult is 800mg 3 times daily. If the symptom is getting better, gradually reduce to 400mg 3 times daily, Maximum dose is 4.8g/day. Administration: May be taken with or without food.
Each tablet/capsule contains:
Piracetam ........ 400mg
Piracetam is a cyclic derivative of gamma-aminobutyric acid (GABA) and belongs to the group commonly referred to as nootropic agents.
The precise mechanism of action of piracetam is not completely established. Its pharmacological effects are considered to involve modulation of neuronal and membrane functions rather than direct sedative or stimulant activity.
Piracetam is rapidly and extensively absorbed after oral administration. It undergoes little or no significant metabolism and is predominantly excreted unchanged by the kidneys.
Its elimination is therefore closely related to renal function.
Contraindications include hypersensitivity to piracetam or related pyrrolidone derivatives, severe renal impairment, and certain conditions associated with cerebral haemorrhage depending on approved labeling.
Use with caution in patients with impaired renal function and those at increased risk of bleeding because piracetam may affect platelet aggregation.
Abrupt withdrawal should be avoided in patients being treated for cortical myoclonus.
Reported adverse effects include nervousness, hyperkinesia, weight gain, somnolence, depression, asthenia and gastrointestinal disturbances.
Management of overdose is primarily symptomatic and supportive. Renal function and the patient's clinical condition should be monitored.
Piracetam has relatively low potential for metabolic drug interactions because it undergoes minimal metabolism. Caution may nevertheless be required with anticoagulant or antiplatelet therapy because of potential effects on haemostasis.
