Domperidone Maleate B.P equivalent to Domperidone ... 10 mg
Domperidone is peripheral dopamine receptor antagonist; antiemetic. It is described chemically a 5-Chloro-1-[1-[3-(2-oxo-2, 3-dihydro-1H-benzimidazol-1-yl) propyl] piperidin-4-yl]-1, 3-dihydro-2H-benzimidazol-2-one. Chemical formula is C22H24ClN5O2 and its molecular weight is 425.9.
Domperidone is dopamine receptor (D2) antagonist which selectively inhibits dopamine at the D2 receptor. It acts principally at receptors in the chemoreceptor trigger zone (CTZ) and also at receptors in the stomach.
Domperidone is rapidly absorbed with peak plasma concentrations approximately 1 hr after oral administration. The absolute bioavailability of oral domperidone is low (approximately 15%) due to first pass hepatic and intestinal metabolism. Domperidone is 91-93% bound to plasma proteins. The plasma t ...
It is contraindicated in patients who have known hypersensitivity to domperidone or domperidone containing products. It cannot be administered if gastric motility causes bleeding, mechanical obstruction or gastrointestinal perforation. It is also contraindicated in patients with prolactinoma and tum...
If an antacid is to be administered concomitantly with antisecretion drugs, it must be given after meals and not administered concomitantly with domperidone. Domperidone is not recommended for long-term treatment. Use with caution in patients with hepatic impairment. In patients with renal insuffici...
Use in pregnancy and lactation: Not recommended in pregnant women and nursing mothers.
Use in children: Because the metabolic and blood brain barrier function are not yet fully developed, the possible occurrence of neurological side effects cannot be totally excluded in infants < 1 year.
Domperidone rarely causes central nervous symptoms, including extrapyramidal symptoms, because of its poor penetration to the blood-brain barrier. Domperidone can induce hyperprolactinema in many patients, resulting in breast tenderness and enlargement, galactorrhea and gynecomastia. Domperidone rar...
Concomitant administration of anticholinergic drugs may inhibit the anti-dyspeptic effects of domperidone. Antimuscarinic agents and opioid analgesics may antagonize the effect of domperidone. Domperidone suppresses the peripheral effects (digestive disorders, nausea, vomiting) of dopamine agonists....
